Abstract:

The Purpose of this experiment is to gain knowledge of how to properly use aseptic techniques to transfer cultures, learning about cultured media and how to distinguish various types of microbial growths as well as what is required for them to grow properly.

Hypothesis:

This exercise will allow me to gain knowledge of how to properly use aseptic techniques, become familiar with basic requirements of microbial growth, cultured media and methods to control their growth. This will further my knowledge and skills in making observations about specimens.

Microbes are found on virtually every surface and in the air, thus using aseptic technique allows the minimization of the cultures exposure to environmental microbes and allowing for a more accurate end result.

E. Describe three common forms of growth that you are likely to see in a broth culture.
1. Pellicle: A mass of organisms floats in or on top of the broth. Smaller masses or clumps of organisms that are dispersed throughout the broth form an even pattern called flocculent. 2. Turbidity: The organisms appear as a general cloudiness through the broth. 3. Sediment: Am ass of organisms appears as a deposit at the bottom of the tube.

F. What is the difference between an aerobe and an anaerobe? Aerobe: an organism requiring oxygen to live. Anaerobe: an organism that can live in the absence of atmospheric oxygen.

G. Describe the difference between facultative and obligate. Facultative: capable of functioning under varying environmental conditions.
Obligate: Able to exist or survive only in a particular environment or by assuming a particular role. H. Which two types of media did you use in this experiment? Why did you need two types of media instead of only one?
The two types of media used to grow the microbial specimens are nutrient medium and MRS medium. The nutrient medium is the standard growth medium used for culturing most microbes, it is consists of heat-stable digestive products of protein and beef
