If we use paper chromatography to separate the pigments in spinach leaves, then we will see several different pigments because plants use other pigments besides chlorophyll a (the green pigment) to capture visible light for photosynthesis.
1. What factors are involved in the separation of the pigments?
a. Some factors that may be involved in the separation of the pigments are size of the molecules, their attraction to the paper, and how attracted they are to the solvent.
2. What type of pigment is mainly used for collecting light energy in the plants, and what makes you think this is the main pigment?
a. I think chlorophyll a (the green one) is the main pigment used for photosynthesis because it is the main pigment that gives the plant its color. It was also the widest band of pigment found, so this indicates that it is the most abundant, and must be important in photosynthesis.
What are the roles of the other pigments found in the spinach leaves?
Chlorophyll is a plant pigment, which gives plants a green color and allows them to absorb light for photosynthesis. Using a quarter extract pigments from leaf cells

Place chromatography paper in graduated cylinder
●
Place chromatography chamber in fume hood and allow chromatograms to develop(Part B)

Turn spectrophotometer to warm up the instrument and set wavelength to 605 nm
●
Setup an incubation area
●
Keep two beakers with chloroplasts on ice
●
Label cuvettes 1-5 respectively
●
Add 1ml phosphate buffer to each cuvette
●
Bring spectrophotometer to zero
●
Obtain unboiled chloroplast suspension and transfer three drops to cuvette 2. Insert inspectrophotometer sample holder.
●
Obtain unboiled chloroplast suspension and transfer three drops to cuvette 3. Insert inspectrophotometer sample holder.
●
Obtain boiled chloroplast suspension and transfer three drops to cuvette 4. Insert inspectrophotometer sample holder.
●
Cover and mix contents of cuvette 5. Insert in spectrophotometer sample holder.
