Lab 1: Microscopy and the Metric System
Purpose
This lab introduced the features of the compound light microscope and reviewed the basics of the metric measurement system.
Materials
1. Tape Measure
2. Scale
3. Thermometer
Part A: Microscopy
1. Eyepiece- the lens the viewer looks through Arm- the frame of the microscope Base- bottom of the microscope where the light is located Objective lens- lens closest to the specimen Fine Adjustment knob- fine tunes the focus on the specimen Coarse Adjustment knob- adjusts the focus on the specimen Stage- where the specimen is placed Stage clips- clips used the hold the specimen in place
2. Commonly, an eyepiece is 10X (meaning magnifies 10 times the size). An images’ magnification is multiplied through the eye piece and the objective lens. So, 4X is equivalent to 40 times magnified, 10X is 100 times magnified, 40X is 400 times magnified, and 100X is 1000 times magnified.
3. Depth of View refers to the area that is in front of and behind the focus point.
4. Field of View refers to the area that is seen when looking through a microscope.
5. To make a wet mount place the sample on the glass slide. Using a pipet, place a few drops of water on the sample. Once the sample is completely submerged in water place the cover glass over the sample. Keeping air out of the sample is very important so place on edge of the cover glass down and slowly lower the other side. Remove any excess water off the slide with a tissue or filter paper.

Part B: Metric System
1. The width of the Campbell and Reece textbook is 23.5 centimeters, which is equal to 235 millimeters.
2. 100 grams = 100,000 milligrams = 100,000,000 micrograms
3. Wooden clothespin- 6.3 grams = 6,300 milligrams = 6,300,000 micrograms
4. The meniscus is the curve that is visible at the surface of a liquid. When taking the measurement of a graduated cylinder, drop to eyelevel and measure at the line that is at the center of the meniscus.
5. Temperature of Skin= 34.4 degrees Celsius Temperature of Air= 22.8 degree Celsius
